Synthesis, Characterization and Anti-inflammatory Activity of 7-azaisatin Derivatives

Objective: Synthesis and evaluation of anti-inflammatory activity of some novel 7-azaisatin derivatives. Methods: Novel Schiff bases of 7-azaisatin (VI) were synthesized by condensation of ethyl-2-(1-benzyl-2-oxo-1,2-dihydro-3H-pyrrolo[2,3b]pyridin-3-ylidene) hydrazine carboxylate (V) with various primary amines. The compound (V) was synthesized by reacting ethylchloroformate with 1-benzyl7azaisatin hydrazone (IV) which was prepared by treating 1-benzylazaisatin (III) with hydrazine hydrate. Oxidation of 1-benzylazaindole gives 1-benzylazaisatin (III). The chemical structures of the synthesized compounds were confirmed by means of IR, 1HNMR, and Mass spectral analysis. The synthesized compounds were investigated for anti-inflammatory activity by carageenan induced paw edema in rats. Results: The results of anti-inflammatory activity showed that most of the compounds exhibited significant activity. Conclusion: Novel Schiff bases of 7azaisatin were prepared, characterized by spectral data and screened for antiinflammatory activity by carageenan induced paw edema in rats. Most of the compounds exhibited promising activity.
